AB5-Type Toxin as a Pentameric Scaffold in Recombinant Vaccines against the Japanese Encephalitis Virus

Japanese encephalitis virus (JEV) is an enveloped icosahedral capsid with a prime neutralizing epitope present in E protein domain III (EDIII). dimers are rearranged into five-fold symmetry of icosahedrons. Cholera toxin B (CTB) and heat-labile enterotoxin (LTB) AB5-type was used as the structural scaffold for emulating pentameric axis EDIII. We produced homo-pentameric EDIII through genetic fusion LTB or CTB E. coli without recourse to additional refolding steps. Harnessing RNA-mediated chaperone further enhanced soluble expression assembly chimeric antigen. The validated by size exclusion chromatography (SEC), non-reduced gel analysis, GM1 binding assay. CTB/LTB−EDIII antigen triggered high antibodies against JEV Nakayama strain after immunization mice. Altogether, our proof-of-principle study creating JEV-protective via both built-in adjuvant posits bacterially recombinant cost-effective alternative conventional inactivated vaccines JEV.
